The isotyping kit contains two cassettes per pouch: one cassette is for determining IgG1, IgG2a, IgG2b and IgG2c, and the other cassette is for determining IgG3, IgM, kappa, and lambda. Both strips contain a control line “C”.

While BALB/c mice express IgG2a, some other inbred mouse strains such as C57BL/6, C57BL/10 and NOD do not have the gene for IgG2a and instead express the IgG2c isotype.

IgG1 and IgG2b bands appear in both strains, but IgG2a only shows up in BALB/c and IgG2c only in C57BL/6. There is no cross-reactivity between IgG2a and IgG2c.
